What Happens When a Telomerase Gene is Mutated?

Abstract

Telomeres protect chromosome ends and prevent genetic loss during DNA replication. The template for synthesizing telomeric DNA is contained in the telomerase RNA, and without this RNA it is predicted that the telomeres will be deregulated. The model organism Aspergillus nidulans, a filamentous fungus, has short, regulated telomeres. In our project, we test the telomerase RNA gene, first identified in our lab, by performing a “knockout” of the six-nucleotide template in living cells. If successful, we expect to see a shortening in telomere length. From this research, we expect to gain novel insight into the regulation and function of telomeres.
